Juan dela Cruz (Coco Martin) is the son of the mortal, Amelia (Mylene Dizon), and is is the latest successor to a line of tagapagbantay. His mother died giving birth to him, leaving the child in the care of Father Cito (Jaime Fabregas), the town's. Unbeknownst to Juan, he also has aswang blood from his demon father, Samuel (Albert Martinez); should the aswang know of this fact, they will force him to join their ranks , Javier The Ultimate Aswang Will Do Anything To Acquire The Earth But Juan Will Stop Him From His Evil Plans.
Juan dela Cruz is an orphan boy who was brought up in the church by a righteous parish priest who guided him towards goodness and faith upon the Lord Jesus Christ, who eventually referred as bossing by Juan. Juan dela Cruz was an innocent child who longs to help people around him but because he had an innate temper and manifestation of superhuman abilities, the prudish townspeople disliked him except the priest whom he regarded as his father. One day, the priest was killed by a thief. Juan ended up in the streets of Quiapo, as a simple, kind and yet tough young man. Unknown to him, he is half-aswang, the son of Samuel Alejandro, the Haring Aswang (King of the Aswangs) and also the prophesied Anak ng Dilim (Child of Darkness). He is fated to rule over the aswangs and take over the human race. However, Amelia, Juan's mother, is a human and a Tagabantay (Guardian), coming from an ancient lineage of defenders using the Bakal na Krus (Iron Cross) to protect the humans from the aswangs.
Juan later joins the Kapatiran (Brotherhood), a group composed of people who seeks to avenge their love ones who were killed by the aswangs. The group helped train Juan into becoming the next Tagabantay. Later the engkantos and diwatas enter the picture to also aid Juan in his duties who seems to have a bigger and important role on the continuing conflict between the aswangs and the humans. The series revolves on Juan's duties as the Tagabantay while continuing learning life lessons and discovering more about himself. The series ultimately debates on destiny, if it is predetermined or a choice. Will Juan choose the side of good despite his dark heritage?
